  • Celecoxib is a highly selective COX-2 inhibitor and primarily inhibits prostaglandin production.
  • Celecoxib is approximately 7.6-fold more selective for COX-2 inhibition over COX-1. This specificity allows celecoxib to reduce inflammation and pain while minimizing gastrointestinal adverse drug reactions (e.g. stomach ulcers) that are common with non-selective non-steroidal anti-inflammatory drugs (NSAIDs).   • Celecoxib shows competitive kinetics on COX-1 (Ki = 10-16 µM). It also displays an initial competitive interaction with COX-2 (Ki = 11-15 µM), followed by a time-dependent interaction leading to potent inhibition and inactivation (Kinact = 0.03-0.5 s-1).
