- Torin1 is a highly potent and selective ATP-competitive mammalian target of rapamycin (mTOR) kinase inhibitor that directly inhibited both complexes, mTORC1 and mTORC2, with IC50 values between 2 and 10 nM in in vitro kinase assays. It prevented cell growth and proliferation to a far greater degree than rapamycin (please see Cat. No. R-5000, Rapamycin). However, these effects were independent of mTORC2 inhibition and were instead dependent on inhibition of rapamycin-resistant functions of mTORC1 that were necessary for cap-dependent translation and suppression of autophagy. These effects were at least partly mediated through mTORC1-dependent and rapamycin-resistant phosphorylation of 4E-BP1. Thoreen, C.C., et al. "An ATP-competitive mammalian target of rapamycin inhibitor reveals rapamycin-resistant functions of mTORC1." J. Biol. Chem. 284: 8023-8032 (2009).
- Torin 1 and Torin 2 are potent antimalarials. Hanson, K.K., et al. "Torins are potent antimalarials that block replenishment of Plasmodium liver stage parasitophorous vacuole membrane proteins." Proc. Natl. Acad. Sci. USA 110: E2838-E2847 (2013).
- Genetic and biochemical evidence obtained using rapamycin or torin 1 and Rheb (Ras homolog enriched in brain) demonstrated that pUL38 prevented endoplasmic reticulum stress-induced cell death independently of its role in mTORC1 activation. Qian, Z., et al. "The human cytomegalovirus protein pUL38 suppresses endoplasmic reticulum stress-mediated cell death independently of its ability to induce mTORC1 activation." J. Virol. 85: 9103-9113 (2011).
